Yes, Markiplier is in a relationship with fellow YouTuber Amy Nelson, who he has been dating since 2015.

Markiplier’s real name is Mark Edward Fischbach, born on June 28, 1989.
Fischbach was born to Clifton M. Fischbach and Sunok Frank. His father, Clifton, was a German American who served in the United States Army, and his mother, Sunok, was a Korean nurse who is now retired.
Fischbach’s parents met when his father was stationed in Korea. After falling in love and getting married, Sunok Frank gave birth to Fischbach at the Tripler Army Medical Center in Honolulu, Hawaii.
Soon after Fischbach’s birth, his family moved to Cincinnati, Ohio, where he was raised in Milford. The future Youtuber went to Milford High School and played trumpet in the school’s marching band.
Fischbach has an older sibling, a brother named Jason Thomas “Tom” Fischbach, an artist and author of the webcomic Twokinds. As he doesn’t like his face being filmed, Tom has only been featured in a few of Markiplier’s videos.
After graduating high school, Fischbach went to the University of Cincinnati to study biomedical engineering. He soon dropped out to pursue his Youtube career.
Sadly, Fischbach’s father passed away after a year-long lung cancer battle on July 4, 2008.
Ten years later, on June 19, 2018, Fischbach’s step-niece Miranda Cracaft departed after being caught in a car accident at 19.
